Plymouth Road begins in Ann Arbor at an intersection with Broadway. It continues into Dixboro and Superior Township, and goes from there into Wayne County where it becomes Plymouth-Ann Arbor Road and then Ann Arbor Trail in Plymouth.
The road is the former route of M-14, before it became a freeway; before that, it was once US-12.
